Default Carb leaks

What causes the carburetor to loose its fuel and needs priming to start, if it has not run for a week or two?

The fuel in the bowl can also evaporate. I pull the choke closed when I shut the engine down.

Most likely the power valve as has been advised.

Be aware that many, perhaps most, new power valves will not seal as they are delivered. I believe most folks get a "good one" from Daytona.

Mike, my 48 started hard when it sat for any extended period. I followed your advice about the choke and it now starts much easier! Thank you.

This can happen even if the power valve is not leaking due to as Kube said, evaporation. Not 100% sure but it seems that the ethanol gas evaporates quicker than regular gas. That's why many add an electric fuel pump to prime the carb.
