Back in 1959/60 my Dad owned a '53 Merc-o-matic, did all 50-53 Merc's have the 255ci motor?

'49-'53 Merc's had the 255 and '51-'53's had the Merc-O-matic's.

Merc-O-Matic was an option,, Lot of 3 speed OD's and some straight 3 speeds,'51 on.

The answer to your question "did all 50-53 Merc's have the 255eng " is yes. that does not mean that after all these years that you are guaranteed to find a 255 c.i. eng. in a Merc. many engines have been swapped out over the years.
