Lenny B - I assume you mean the felt channels that the windows ride up and down in. If so, whether you HAVE to remove the glass depends on what kind of car (as Brent pointed out). On the steel door cars the felts are held to the doors by clips, and you can (with difficulty) remove them by sliding them out after releasing the clips. On the wood frame doors (the fordors and Town Sedans) the felts are nailed to the wood frame, and you need to get the window out of the way to get to some of the nails.
If you are lucky and have a coupe or pickup, the top of the door unscrews and the window lifts out the top (after releasing the regulator). Either way, likely the easiest would be to just pull the windows out however you have to do it, that way if you have to put some muscle into the job you don't run the risk of damaging the glass.
