I went and looked in the intake valley again. I think that "hole" is just the way the photo looks. I found no hole above the main cam gear either from the inside or outside of the block.

Thanks, Fred. That means I must've drilled the hole in my 221. Still can't recall why I did it 50+ years ago --- must be having yet another senior moment. 8^)
